HIV-1 GROUP M CLADES INFECTING PERSONS IN RURAL VILLAGES IN CAMEROON

OBJECTIVE:
To identify the HIV-1 group M subtypes infecting persons living in rural villages in five provinces of Cameroon.
METHODS:
As part of a study of HIV-1 prevalence in rural villages in Cameroon, five provinces (Center, East, South, West and South West) were visited during the period April 2000 to October 2001. After a verbal consent, socio epidemiological data were collected followed by a collection of 10ml of blood in an EDTA tube by veino-puncture. After centrifugation, aliquots of sera were stored at -20o C for RNA extraction. RNA was extracted and env and gag gene were amplified. Infecting subtypes were determined using env and gag Hetero Mobility assay (env & gag HMA) or sequencing followed by phylogenetic analysis.
RESULTS:
Seventy-nine plasma were analyzed so far. The HIV-1 group M clades identified included A, D, F (F2), G, H and C. The CRF strains identified were CRF02_AG-like, CRF01_AE-like and CRF11_cpx. 57% (45) of the group M infections were clade A or CRF02_AG-like as subtype by env and gag. Of the remaining group M infections, 14% (11) were either A or CRF02_AG-like in recombination with other clades; 25% (20) were infections that were entirely non-A or non-CRF02_AG- like and 4% (3) were CRF11_cpx partially in one gene (gag) or totally in both (env and gag). Three new intersubtype recombinant infections, H/G, A/F2 and H/A were identified.
CONCLUSIONS:
This study suggest that the HIV-1 diversity in rural villages in Cameroon is at least as broad as has been observed in major cities of Cameroon and that multiple HIV-1 group M subtypes are infecting persons living in countryside of Cameroon.
