Abstract
Molecular epidemiology of HIV-1 in Cameroon: decrease in the proportion of mosaic CRF_02.AG from west to east Cameroon

Background: Numerous studies have provided a picture of the HIV-1 group M distribution among samples taken from various studies/towns in Cameroon. All show the high prevalence of HIV-1 subtype A and CRF02_AG.
Objectives: In this study we carried out the molecular epidemiological studies on HIV-1 and its relating viruses in west and east Cameroon. By such monitoring surveys, we may be able to clarify how and to which direction the virus is changing genetically.
Material and methods: We analyzed 107 HIV-1 positive samples, from rural eastern (n= 57) and western (n=50) Cameroon. Genomic DNA was extracted, PCR amplified, cloned and sequenced. Analysis software utilized were as follows; PAUP*, PALM and MEGA for sequence analysis. A phylogenetic tree was constructed by the neighbor-joining method, and its reliability was estimated by 1000 bootstrap replications.
Results and discussion: Our data in rural eastern Cameroon highlight a relatively low prevalence of CRF02_AG (29.8 and 35.0%) and relatively high prevalence of CRF11_cpx (21.4 and 17.5%) based on gag-p24 and env-C2V3 analyses, respectively. So far tremendous amount of sequence data have been accumulated from western Cameroon in our data base (Ndembi et al. unpublished data and Tebit et al. ARHR 2002). The new sequences were distributed among subtype A, G and CRF02_AG strain. The majority of the new isolate clustered with CRF02_AG (42.1% and 72.0% ; p< 0. 05) based gag-p24 and env-C2V3 analyses.
Conclusions: In the east of Cameroon, Chad, relatively low prevalence of CRF02_AG (13.1%) and relatively high prevalence of CRF11_cpx (13.1–18.0%) and subtype D (18.7%) compared with those in neighboring countries such as Nigeria, where CRF02_AG is reported to be predominant . These findings highlight a decrease in the proportion of mosaic CRF02_AG from west to east Cameroon.
