Abstract



New HIV inhibitors detected among drugs of different (non-anti-HIV) pharmacologic effect

Background: To search for antiretroviral drugs among agents, widely applied for the treatment of somatic and infectious diseases, rather than AIDS. In case of success such drugs could be included in the scheme of highly active antiretroviral therapy (HAART).

Methods: The investigations were carried out on the continuous lymphoblastoid cell lines CEM.SS, Jurkat-tat3, Molt4/8 and MT-4. The rapid/high strain HIV-1zmb and its AZT-resistant variant, isolated from the virus carrier, were used. The therapeutic activity (the tested agent was added simultaneously with the virus) as well as the prophylactic activity (the virus was placed on tissue culture 2 hours after the tested agent) was checked. The following tests were used for trials: a) pharmazane (MTT) test; b) indirect immunofluorescence test; c) staining of infected cells with trypane blue stain. The last two tests were used only in case when MTT-test was positive.

Results: A total of 307 drugs prescribed as a rule per os for the treatment of vascular, neurologic and infectious diseases were tested. Among them 11 drugs, including 6 ones with high antiviral activity, were revealed. Only three of them – Tardyferon, Xantinol Nicotinat and Pentoxyphillinum received a detailed investigation because they had moderately marked side effects, were at a reasonable price, inhibited replication of AZT-resistant HIV-1 strain, showed synergism in combination with AZT and were compatible with each other.

Conclusions: Two active in vitro medical drugs (vasodilative antiaggregative drug Xantinol Nicotinat and hemopoietic agent Tardyferon) require clinical trials (I-II phases) in order to determine the effective and safe dose for HIV-infection. This dose for Pentoxyphillinum makes up 1200 mg per day. In case of positive outcome of the experiment they could be used as additional components of HAART.
